The standard dosage of linaclotide for irritable bowel syndrome with constipation is 290 mcg once a day. Linaclotide is taken in the morning, at least 30 minutes before your first meal of the day. Talk to your healthcare provider if you have bothersome side effects on this amount. It is possible that your dosage could be reduced.
 

Linaclotide Dosing Guidelines for Chronic Idiopathic Constipation

The standard dose of linaclotide for chronic idiopathic constipation is 145 mcg once a day. In clinical studies, doses of 290 mcg a day did not work any better than the 145 mcg a day dose. You will usually take your dose in the morning, at least 30 minutes before your first meal of the day.
